Effects of Benzocaine, Clove oil, and Tricaine Methanesulfonate (MS-222) on Plasma Cortisol in Crucian Carp (Carassius auratus)

Abstract
Benzocaine, tricaine methanesulfonate (MS-222), and clove oil have been widely uesd for anesthesia in fishes. To determine the effects of these three anesthetics on the secondary stress indices, changes in plasma cortisol level were monitored in healthy crucian carps (Carassius auratus). 36 fishes were divided into four groups, and blood was collected before anaesthesia (control group), immediately after anesthesia on different times(stage 1; sedation, stage ¥± ; pre-anaesthesia and stage ¥² ; anaesthesia). As compared with control group, tricaine methanesulfonate gradually increased plasma cortisol level with increasing stages. Meanwhile, benzocaine and clove oil decreased plasma cortisol level with increasing stages. Particularly, clove oil dramatically decreased plasma cortisol level at stage ¥², which is almost twice lower than the control group. These results suggest that benzocaine and clove oil effectively reduced plasma cortisol levels. Therefore, clove oil can be an effective anesthetics with stress-reducing capability.
